EVALUATION OF SNP MARKERS ASSOCIATED WITH QUALITY OF MEAT IN CREOLE AND CROSS-BORDER POPULATIONS
Abstract
The polymorphism of five SNP markers associated with meat quality, DGAT1, TG1, MSTN80, MSTN42, and MSTN99, was determined by NGS sequencing. The polymorphism of DGAT1 (g.1802265A>G) is located on chromosome 14 of the bovine genome; Thyroglobulin, TG1, (g.9487659A>G) located on the same chromosome, and three variants of the Myostatin gene, MSTN80 (g.6213980A>C), rs110065568; MSTN42 (g.6215942T>C), rs137528458, and MSTN99 (g.6218499A>G), c.938G>A located on chromosome 2. SNP analysis was performed using the sequencing panel. Library preparation was carried out following the manufacturer's workflow and sequenced using bridge amplification and sequencing by synthesis on a MISEQ system. The most informative markers were DGAT1 and MSTN42, with DGAT1 showing the highest Hob and He values of 0.424 and 0.430 respectively, and the lowest Hob and He values observed in MSTN42 with 0.322 and 0.328 respectively. Polymorphism was determined in the genotypes subjected to this study; however, fixation of TG1, MSTN80, and MSTN99 SNP alleles was observed in all genotypes. The MSTN42 marker is reported for the first time in native and cross-border breeds and genotypes in Panama. The results suggest the feasibility of genetic improvement for quality meat production, particularly in Guaymí and Guabalá native cattle. Increasing populations is required to improve the precision of the markers used in this study.
